Bayesian analysis of minimal model under the insulin-modified IVGTT

چکیده
A Bayesian analysis of the minimal model was proposed where both glucose and insulin were analyzed simultaneously under the insulin-modified intravenous glucose tolerance test (IVGTT). The resulting model was implemented with a nonlinear mixed-effects modeling setup using ordinary differential equations (ODEs), which leads to precise estimation of population parameters by separating the interand intra-individual variability. The results indicated that the Bayesian method applied to the glucose-insulin minimal model provided a satisfactory solution with accurate parameter estimates which were numerically stable since the Bayesian method did not require approximation by linearization.

Bayesian hierarchical approach to estimate insulin sensitivity by minimal model.
We adopted Bayesian analysis in combination with hierarchical (population) modelling to estimate simultaneously population and individual insulin sensitivity (SI) and glucose effectiveness (SG) with the minimal model of glucose kinetics using data collected during insulin-modified intravenous glucose tolerance test (IVGTT) and made comparison with the standard non-linear regression analysis. متن کاملOverestimation of minimal model glucose effectiveness in presence of insulin response is due to undermodeling.
Glucose effectiveness is an important determinant of glucose tolerance that can be derived from minimal model analysis of an intravenous glucose tolerance test (IVGTT). However, recent evidence suggests that glucose effectiveness is overestimated by minimal model analysis. Here we compare a new model-independent estimate of glucose effectiveness with the minimal model estimate by reanalyzing pu...
متن کاملModeling glucose and free fatty acid kinetics during insulin-modified intravenous glucose tolerance test in healthy humans: role of counterregulatory response.
Insulin administration during insulin-modified intravenous glucose tolerance test (IM-IVGTT) can induce transient hypoglycemia in healthy insulin-sensitive subjects. This triggers counterregulatory reflex (CRR) responses, which influence the kinetics of glucose and nonesterified fatty acids (NEFA), and undermines the accuracy of mathematical modeling methods that do not explicitly account for C...
متن کاملMinimal model SGoverestimation and SIunderestimation: improved accuracy by a Bayesian two-compartment model.
The intravenous glucose tolerance test (IVGTT) single-compartment minimal model (1CMM) method has recently been shown to overestimate glucose effectiveness and underestimate insulin sensitivity. Undermodeling, i.e., use of single- instead of two-compartment description of glucose kinetics, has been advocated to explain these limitations. We describe a new two-compartment minimal model (2CMM) in...
متن کاملMultiscale modelling of insulin secretion during an intravenous glucose tolerance test.
Dysfunctional insulin secretion from pancreatic β-cells plays a major role in the development of diabetes. The intravenous glucose tolerance test (IVGTT) is a widely used clinical test to assess β-cell function. The analysis of IVGTT data is conveniently performed using mathematical models, which need to be fairly simple to enable parameter identifiability (minimal models), but should at the sa...
